Importante d\u00e9couverte \u00e0 Philadelphie"},"content":{"rendered":"\n<p>En 1960 \u00e0 Philadelphie, les scientifiques Peter Nowell et David Hungerford ont jet\u00e9 les bases du d\u00e9veloppement du Glivec en d\u00e9montrant qu&rsquo;un fragment du chromosome 22 faisait d\u00e9faut sur 95% des patients atteints de LMC. Ce chromosome mutant fut d\u00e9sign\u00e9 par l&rsquo;endroit o\u00f9 il fut d\u00e9couvert: le chromosome de Philadelphie (illustration 5.1). C&rsquo;\u00e9tait la premi\u00e8re fois que des chercheurs parvenaient \u00e0 trouver un lien direct entre une modification g\u00e9n\u00e9tique et la formation d&rsquo;un cancer.<\/p>\n\n\n\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" src=\"https:\/\/biotechlerncenter.interpharma.ch\/wp-content\/uploads\/sites\/4\/2020\/02\/grafik5-1_fr.jpg\" alt=\"\" class=\"wp-image-4196\"\/><figcaption>Illustration 5.1: Formation du chromosome de Philadelphie<br>\u00a9 Interpharma<\/figcaption><\/figure>\n\n\n\n<p>Plus d&rsquo;une dizaine d&rsquo;ann\u00e9es plus tard, on d\u00e9couvrit que le fragment d&rsquo;ADN manquant sur le chromosome 22 avait migr\u00e9 sur le chromosome 9. Il a fallu \u00e0 nouveau une dizaine d&rsquo;ann\u00e9es pour que les chercheurs comprennent que cette translocation \u00e9tait \u00e0 l&rsquo;origine de la LMC. Par la translocation des segments d&rsquo;ADN, deux g\u00e8nes vont nouvellement fusionner, ce sont les g\u00e8nes&nbsp;<em>Bcr<\/em>&nbsp;et&nbsp;<em>Abl<\/em>. (Remarque: afin de pouvoir diff\u00e9rencier les g\u00e8nes des prot\u00e9ines, il fut d\u00e9cid\u00e9 unanimement que les g\u00e8nes seraient \u00e9crits en italique et les prot\u00e9ines en caract\u00e8res normaux.). Les deux g\u00e8nes devinrent soudainement voisins et form\u00e8rent un oncog\u00e8ne, un g\u00e8ne g\u00e9n\u00e9rateur de cancer.<\/p>\n\n\n\n<p>La nouvelle association de&nbsp;<em>Bcr<\/em>&nbsp;et&nbsp;<em>Abl<\/em>&nbsp; va former une nouvelle prot\u00e9ine appartenant \u00e0 la classe des tyrosine-kinases. Ces prot\u00e9ines peuvent conduire les signaux de l&rsquo;ext\u00e9rieur \u00e0 l&rsquo;int\u00e9rieur de la cellule: de ce fait, la cellule sera renforc\u00e9e \u00e0 l&rsquo;encontre des influences ext\u00e9rieures ou pourra entrer en contact avec les cellules avoisinantes.
